Description

The PacBio RS II sequencer uses single molecule, real time (SMRT) sequencing technology which takes advantage of immobilized DNA polymerase/template complex nested in thousands of small wells (called zero-mode waveguides). These wells can measure changes in light emission arising from the processing of nucleotides that are each linked to a different colored fluorophores. As each nucleotide is incorporated by the polymerase through natural DNA synthesis, the fluorescent emission from the fluorophore release are detected in parallel by the wells, generating a large amount of sequence data. The RS II system reports an average read length of over 10,000 bp and can reach read lengths of 60,000 bp. Each cell generates around 55,000 reads. The system features the flexibility of running between 30 minutes to 6 hours per SMRT cell and can run between 1 to 16 cells at a time. Other features include a touchscreen interface, a workdeck, and robotic workflow management.
